Question : DLL kill

How to kill Dlls in Windows 2000. Any techniques, tools available. I need to program/write a DLL  and every time I write a new DLL file, I have to delete the old one from WINNT/System32, which I cannot do, since it is in use. I can get around this by restarting my machine, but u know .... this same old egg and hen, which came first, story !!!!
