Click once is per user/manifest based deployment. Makes updates easy.
I don't believe it allows for a silent mass first install.
I don't know your specific setup so this may be farfetched but couldn't you just send out an e-mail to all those users with the link to the install?
If you need a silent install you may need to use something else. :-(
